There is no real, comprehensive guide on the 'setting', but it is analogous to Earth in every way, containing the same cities and historical events, just with a supernatural sort of twist, including things like Vampires and Stand Users.
Personality:
Dio's personality is best summed up in describing him as a megalomaniac with antisocial personality tendencies. To start, he has delusions of being 'above' other people, none of which directly stemmed from his turning into a vampire, though the transformation did manage to exacerbate the thoughts, really cementing them in his mind as he placed himself 'above' humanity. But even in youth, he believed that he was 'better' than others. Stronger, faster, smarter, and more worthy, and he was more than willing to prove himself to these ends, usually to his adoptive brother's dismay. He has a certain sense of pride in everything that he does, from gloating about learning to fight from the slums to showing off his intelligence. He wins a fight against Johnathan, his adoptive brother, in a boxing match quite easily, and then continues to show off 'his technique' to the kids who had been watching and he bests Jojo in grades as well, graduating as valedictorian for the sole reason that he wished to break his brother's spirit.
And he certainly DOES show off, more than is necessary. Dio is, like much of Jojo's Bizarre Adventure, extraordinarily theatrical, which comes along with being the main villain for most of the series. Everything he does, says, and wears has a bit of an extra flair when it counts. In fact, he does have a bad habit of reminding people of his name in normal conversations with his manner of speech, as if he were a character in a play rather than someone acting naturally. His style of dressing is also based in this need to stand out, as well as the comfort he takes in his body, as he tends to wear revealing or skin-tight clothing, sometimes with a cape or some other long, flowing accessory, and nearly always provocative in nature, and he tends to pose his body in sometimes strange and always exceedingly confident ways.
Speaking of a provocative nature, Dio is canonly bisexual(though this is mostly Word of God, and no solid examples of his interest in men have been given aside from his manner of dress and his comfort level in draping himself over his male 'friends', up to and including sticking his leg in between the legs of one of them or lying on a bed with them while half-dressed) and has been described as 'throwing himself at either gender'. He is a pleasure seeker, and has no problem in indulging himself in any facet. He likes to partake in sexual activities, drinking alcohol, and other such things that would be labelled 'risky behavior'(though it is unlikely that he partakes in illegal substances, as while he is indulgent, he is somewhat vanilla in his indulgences). In fact, he does have alcohol problems at times, often drinking when he is stressed, despite it being something that disgusts him because of his alcoholic father. It's something he does have trouble controlling.
Dio is also known for his ambition and ruthlessness. He isn't concerned in the least with social mores or ethics when it comes to getting what he wants, and he will usually go with the most extreme method to meet his goals first, never doing things in half-measures. He's not afraid to resort to the most gruesome violence that he can think up, so long as it will get the effect that he prefers, and of course, he DOES prefer violence just for the sake of it at times.
This doesn't mean, however, that he has a hard time interacting with most people. If one is a subordinate to him, or even if they are simply unrelated to his ambitions, Dio can appear to be downright friendly and charming. Despite his villainous nature and his mile-long sadistic streak, Dio has developed in his century of life the ability to truly appreciate the company of others, and he can, in fact, form (somewhat)meaningful relationships with people, though they are also subject to his manipulation and deception. He can be perfectly pleasant with these people, enjoying spending time with them, even if it is just reading a book alongside them or waxing philosophical about religion and reality. They are important to him in such a way that he has 'invested' in them, and he values loyalty above anything and often tests the loyalty of these 'friends', including when he asked Vanilla Ice to give him his blood and when he asked Pucci why he did not simply take his Stand(Pucci's ability was to be able to take the Stands and memories of other people by taking a disc out of their heads) and then forcing his fingers into his hand and allowing him to even hold the disc to see if he would betray him. Both of these two 'passed' their tests, which endeared him to them, though if the trust was ever broken, he would have no regrets about cutting them down.
Dio has a love of intellectual pursuits as well, often being seen reading a book, which can be of any number of subjects. He also went to a prestigious academy, where he had intended to graduate as top of his class in law, though he never got to graduate due to the whole 'trying to kill his father, being found out and turning into a vampire' thing. Even so, his love of knowledge does not stop at old-fashioned knowledge or the liberal arts. He is impressed and fascinated by the technology of today, and has not had many issues in keeping up with it, being willing and able to use television, automobiles, and quite likely telephones and other such modern conveniences. He even mentions watching television at some point(a Micheal Jackson concert, at that), and he shows particular interest in automobiles(he touches one in a manner that is usually taken as suggestive as he mentions that 'in his youth, there were only carriages). He even interjects a simile about an airplane stewardess serving a first-class passenger when he orders a woman to fetch him his leg, and is known to travel the world quite a bit, so it's fair to assume he takes red-eye flights often.
He is not, however, the calm, cool and intellectual villain that he likes to portray himself as. He certainly does fake it well, when things are going his way, but he is prone to panic when he is cornered, and does things that he later feels ashamed for(though his pride would never allow him to admit his shame). He has cried after being punched by his adopted brother, attempted to reason with him that 'he was only acting out because of his upbringing' and lied saying that he would turn himself in to the police, and even tried to offer his brother(then his enemy) immortality in order to get him to release him so that he would not be killed by an impending boat explosion. In his older years, he is more cautious, and so he gets less opportunities to show this panicked state, but in the end, after he's nearly defeated and he realizes that he can't win he does lose his composure again, showing fear and attempting to run away.

Basically a bara vampire. There is a lot of good fanart, but you have got it about right if you think of a muscular guy at 6'4" with blond hair and red eyes who likes to wear skin-tight or revealing clothes with some really strange additions because Jojo.

Jojo-verse vampires have some interesting abilities, including: heightened strength/speed/senses, drinking blood with tentacles/blood vessels in their fingers, the ability to regenerate even more quickly than fire can consume their body(though granted after a point, they can only regenerate by drinking more blood), the ability to freeze their own body and whatever they touch, hypnotize people by sight, plant 'buds' in peoples' minds to control them from a distance(though it weakens the person who is controlled considerably), and they can manipulate their body pretty completely(utilized by Dio when he cuts his own head off and uses the blood vessels to get around in his final battle with Johnathan, as well as him being able to pressurize his bodily fluids and shoot them like a laser from his eyes).
He also has an aversion to direct sunlight and sunlight-based powers. As in a 'vaporizes in sunlight' type aversion, though I understand(from reading the FAQ) that this will be downplayed in-game to a more deathless weakness and lethargy in sunlight. He is not affected by garlic, crosses, running water, or the 'rule of courtesy'(when a vampire cannot come in uninvited).
In addition to his vampiric powers, Dio develops a Stand in Stardust Crusaders.

A Stand is a spiritual manifestation of power that usually(but not always) takes on a humanoid appearance. A Stand cannot usually be seen by anyone but a Stand user, and cannot be affected by anyone but a Stand User(this will be a bit different in-game than in canon, due to different universes), and any damage to the Stand is reflected on it's User as well as vice-versa.
Dio's Stand is called The World. It can go up to 10m away from it's user and is very fast and powerful, using it's fists/legs to fight. Besides that, The World has the ability to stop time for up to 11 seconds.

So he might have said a few things, perhaps, that should not have been said, but the damage is done now, and there was no use in trying to pretend he was not a vampire. Anyone seeing it handwritten as it was would notice remarkable penmanship, his letters flowing but simple. Not quite cursive, but certainly not the print of someone who was born in any recent century. At least, not unless they were practicing calligraphy.]
It has come to my attention that I, Dio, have shared information on this 'network' of a sensitive nature. I do not wish to recant my statement, as I do have an aversion to sunlight and require blood to survive.
However, I am certain that we can all agree that this is simply a minor setback. I have no need to kill anyone for my sustenance as it is, and it is my belief that we will survive more easily if we should work together in this situation as opposed to raising our proverbial torches and pitchforks against each other.
I, Dio, am willing to let the past remain in the past as long as we all are ready to move on and not hold something like my state of undeath draw us apart.
[Yeah, that was convincing enough, right? He didn't think that could be misconstrued as 'I will kill everyone who says a cross word about me', though it might be viewed as a desperate attempt to simply not starve to death. Ugh.

A shame, a crying shame that he had to be awake during the daytime. Dio had been ecstatic at first to learn that the sun did not actually vaporize him immediately, especially since shelter here had been scarce at best and the people were diurnal, though it did cost him much of his strength and energy.
As it was, he spent the time that he wasn't actively on the move lying down in the sand and drawing circles with his finger. He'd prefer to be napping.
With a hand on his chin, his red eyes from the ground up to the sky as he gave a sigh. Was this the 'heaven' he'd been working towards? It was possible, though unlikely. He had not predicted such an outcome from his research, but then... was there ever a way to tell how things would go without a test? He looked over his shoulder as he summoned his Stand, The World, the golden humanoid form appearing behind him silently, arms crossed. Yes, The World existed still, though it was supposed to have been destroyed in the 'ritual'. Even so, he had mentioned it possibly being only temporary, so the verdict was still out on that one. This was going to require some more studying of the situation to truly understand.
Well, if it was indeed his Heaven, there would be no Joestars, and he'd not yet seen or sensed any, which was a great sign! Dio did, however, need to have a goal in mind, in that case. A man without ambition was hardly a man at all! Pulling together his strength, sapped as it was by sunlight, he pushing himself up to his feet, the Stand fading with the sandstorm.
"It seems that I, Dio, will need to find more subordinates to replace the ones I have lost in the changing of this reality", he said mostly to himself, looking around. These people here were good enough as entertainment, certainly, but to truly make the most of this new life, he would need them not to be his peers, but his underlings. Every one of them. Yes, they would bow to him given enough time. Who could deny him for long anyway? He was charismatic, beautiful, and powerful, and most importantly, this was HIS world. He'd created it, and there was not a single person to deny his right to rule it. Especially since his old nemesis, the Joestar bloodline, had finally perished.
A wicked smile touched his lips as he decided that this would simply be another test for Dio. His final test in strength and intelligence, and he was confident in his ability to ace it.
